Mesenchyma stem cells (MSC) can be isolated from various organs of the body such as bone marrow, adipose tissue, umbilical cord, placenta, hair follicle, gingivaland deciduous teeth. After the use of appropriate culture media, mesenchymal stem cells are able to differentiate into various tissues, including osteocyte, chondrocyte, adipocyte, myocyte, hepatocyte and neuron cells. Given their wide differentiation potential and self-renewal capacity, MSCs have been considered a promising candidate for cell-based therapy and tissue engineering.

 
(A) CD marker  identification of mesenchymal stem cells
Mesenchymal stem cells express their specific pattern of CD markers. Using flow cytometry to assess CD marker expressions , it was found that CD44 and CD90 are positive markers, rather than CD14, on mesenchymal stem cells.
 
 
(B) Assessment of MSC differential activity
Besides CD marker identification, assessment of MSC differential activity is also an important issue for MSC characterization. After a period of time treatment for MSC by appropriate induction medium, these cells would express specific markers related to its differentiation lineage. For example, Oil red staining is used to detect adipocyte differentiation, ALP and Alizarin Red staining is for osteocyte differentiation, as well as nestin, GFPA and MAP staining is for neuron differentiation.
